Aqueous biphasic systems (ABS) composed of ionic liquids (ILs) are promising platforms for the extraction and purification of proteins. In this work, a series of alternative and biocompatible ABS composed of cholinium‐based ILs and polypropylene glycol were investigated. The respective ternary phase diagrams, tie‐lines, tie‐line lengths and critical points were determined at 25°C. The extraction performance of these systems for commercial bovine serum albumin (BSA) was then evaluated. The stability of BSA at the IL‐rich phase was ascertained by size exclusion high‐performance liquid chromatography and Fourier transform infrared spectroscopy. Appropriate ILs lead to the complete extraction of BSA for the IL‐rich phase, in a single step, while maintaining the protein's native conformation. Furthermore, to evaluate the performance of these systems when applied to real matrices, the extraction of BSA from bovine serum was additionally carried out, revealing that the complete extraction of BSA was maintained and achieved in a single step. The remarkable extraction efficiencies obtained are far superior to those observed with typical polymer‐based ABS. Therefore, the proposed ABS may be envisaged as a more effective and biocompatible approach for the separation and purification of other value‐added proteins. 相似文献

Sample homogenization is an essential step for genomic DNA extraction, with multiple downstream applications in Molecular Biology. Genotyping hundreds or thousands of samples requires an automation of this homogenization step, and high throughput homogenizer equipment currently costs 7000 euros or more. We present an apparatus for homogenization of individual Drosophila adult flies in 96-well micro-titer dishes, which was built from a small portable paint-shaker (F5 portable paint-shaker, Ushake). Single flies are disrupted in each well that contains extraction buffer and a 4-mm metal ball. Our apparatus can hold up to five 96-well micro-titer plates. Construction of the homogenizer apparatus takes about 3–4 days, and all equipment can be obtained from a home improvement store. The total material cost is approximately 700 euros including the paint-shaker. We tested the performance of our apparatus using the ZR-96 Quick-gDNA™ kit (Zymo Research) homogenization buffer and achieved nearly complete tissue homogenization after 15 minutes of shaking. PCR tests did not detect any cross contamination between samples of neighboring wells. We obtained on average 138 ng of genomic DNA per fly, and DNA quality was adequate for standard PCR applications. In principle, our tissue homogenizer can be used for isolation of DNA suitable for library production and high throughput genotyping by Multiplexed Shotgun Genotyping (MSG), as well as RNA isolation from single flies. There is growing evidence of restoration success for wetland plant communities. However, little research has been done on the associated invertebrate community. We test whether restoring plant communities after peat extraction is sufficient for restoring the taxonomic and functional composition of beetle communities. We monitored taxonomic and trait‐based community metrics for beetle assemblages on restoration islands that were up to 13 years old and compared these with the adjacent “target” undisturbed peat bog. Recovery of beetle abundance, species richness, and trophic structure on the islands was remarkably rapid (i.e. within a decade) and converged on that of the undisturbed peat bog within 13 years after restoration commenced. In contrast, small, native, and poor‐dispersing taxa were persistently less abundant on the islands than in the undisturbed peat bog, causing persistent differences in species composition, even on the oldest islands. These poor‐dispersers probably need assistance to reach the islands and possibly ongoing intervention to allow them to survive there. Our findings emphasize the potential for functional trait analysis to reveal barriers to full restoration of insect community composition. 相似文献

The lipid composition of microbial communities can indicate their response to changes in the surrounding environment induced by anthropogenic practices, chemical contamination or climatic conditions. A considerable number of analytical techniques exist for the examination of microbial lipids. This article reviews a selection of methods available for environmental samples as applied for lipid extraction, fractionation, derivatization and quantification. The discussion focuses on the origin of the standard methods, the different modified versions developed for investigation of microbial lipids, as well as the advantages and limitations of each. Current modifications to standard methods show a number of improvements for each of the different steps associated with analysis. The advantages and disadvantages of lipid analysis compared to other popular techniques are clarified. Accordingly, the preferential utilization of signature lipid biomarker analysis in current research is considered. It is clear from recent literature that this technique stays relevant – mainly for the variety of microbial properties that can be determined in a single analysis. 相似文献

基于2000—2012年张掖市混合型水资源投入产出模型,运用改进的假设抽取法,分析了张掖市6部门水资源关联效应,揭示了水资源在行业间的消耗规律,为调整产业结构提供有利的参考。研究结果表明:(1)各部门水资源直接消耗量与满足自身所需的水资源量不对等,种植业的水资源直接消耗量和纵向集成消耗量均为最大,且其纵向集成消耗水量小于直接消耗量,是张掖市经济系统中真正的水资源净输出部门。(2)种植业的内部效应和复合效应均最大,对自身的依赖性极强。服务业的净后项关联最大,对其他部门的依赖程度最高。(3)水资源在各部门之间发生了转移,种植业是张掖市经济系统中最大的水资源供给者,服务业是各部门中最大的受水者,通过中间投入的方式,由种植业到服务业的路径是最大的水转移途径,而建筑业是"纯"输入部门。(4)2000—2012年间,各部门的内部效应、复合效应、净前项关联和净后项关联均变化显著,进一步反映了产业部门水资源利用的动态关联。 相似文献

近年来我国城市化进程不断推进,不仅体现在城市面积上的增长,也体现在建筑物高度的增长。高度增长一方面能尽量克服城市土地资源匮乏的瓶颈,另一方面为优化城市结构及城市功能做出贡献。在城市遥感研究领域,对于城市建筑物高度的提取也成为研究的重点。城市建筑物高度的估计与测量,已成为城市规划和扩张、城市灾害风险预警与评估的重要参数,同时也为数字城市三维模型的建立提供了基础测绘资料。分别基于光学遥感影像、高分辨率SAR(Synthetic Aperture Radar)影像以及光学遥感影像与高分辨率SAR影像的融合三方面,全面阐述城市建筑物高度的提取方法,并比较两类影像在提取建筑物高度的优劣势,通过回顾早年研究方法,逐步引入近年来新的发展趋势。 相似文献
